Chromogenic Coliform Agar (pH 6.8 ± 0.2) is a ready-to-use, selective and differentiating chromogenic agar for the detection and determination of bacteria count of Escherichia coli (E. coli) and coliform bacteria in water samples with minimal bacterial flora in the background, in accordance with ISO 9308-1 and the new German drinking water regulation.
The combination of two chromogenic substrates allows the simultaneous detection of coliform bacteria and E. coli. The chromogenic Salmon®-GAL substrate (6-chloro-3-indolyl-β-D-Galactopyranoside) is cleaved by the ß-D-Galactosidase, characteristic for coliforms, and results in a pink-red colouring of positive colonies.
The cleavage of the chromogenic X-Glucuronide substrate (5-bromo-4-chloro-3-indoxyl-β-D-Glucuronide) by the ß-D-Glucuronidase, characteristic for E. coli, results in a blue colouring of positive colonies. The colonies turn dark blue violet as E. coli cleaves both Salmon®-GAL and X-Glucuronide. This makes it easy to distinguish them from the other coliforms which appear in a pink-red. The Tryptophan contained in the medium improves the indol reaction for the further confirmation of E. coli, thus increasing the detection reliability.
